Abstract

Antiplatelet aggregation effect of garlic (Allium sativum L.) water extract, turmeric (Curcuma domestica Val.) ethanolic extract and its combination to Swiss Webster mice has been investigated. The test substances given orally during the period of 28 days. Antiplatelet aggregation effect was tested by measuring bleeding time, coagulation time, anddecreasing plasma absorbance to observe platelet aggregation activity before and after addition of ADP as platelet aggregation inducer. Based on the inhibition of plasma abdorbance decrease, turmeric extract at a dose of 100 mg/kg body weight, garlic extract at a dose of 100 mg/kg body weight, combination of turmeric at a dose of 100 mg/kg body weight and garlic extract at a dose of 100 mg/kg body weight as well as combination of turmeric at a dose of 50 mg/kg body weight and garlic extract at a dose of 50 mg/kg body weight showed the effect of platelet aggregation inhibition of 73.67 ± 10.30% (p=0.021); 74.67 ± 25.04% (p=0.018); 77.43 ± 8.09% (p=0.018); and 57.00 ± 23.87% (p=0.047) respectively. The test substances above respectively increased bleeding time of 473.08 ± 56.12% (p<0.000); 427.53 ± 66.89% (p<0.000); 515.73 ± 114.47% (p= 0.001); 481.73 ± 116.51% (p=0.001). Increasing coagulation time only showed by combination of turmeric and garlic extract at a dose of each 100 mg/kg body weight and also of each 50 mg/kg body weight with percentage of 46.67 ± 21.89% (p=0.014) and 40.84 ± 25.80% (p=0.018).Key words : Antiplatelet aggregation, turmeric, garlic, ADP.

Abstract

Oral drug absorption administered is highly influenced by dissolution rate, especially for poorly and insoluble drugs. Tween 80 and polyethylene glycol 400 (PEG 400) as surfactant an increase drug solubility by means of a micelle forming mechanism. The aim of the study wasto know the influence of addition and variation of Tween 80 and PEG 400 on the absorption of piroxicam from rat intestinal lumen in situ. Piroxicam solution in pH 7.5 phosphate buffer was prepared with the same concentration in three kinds based on the addition concentration of Tween 80 and PEG 400, they were 1.0, 1.5 and 2.0 % respectively. The male rats of 130-180 grams body weight fasted were for 24 hours before treatment. The rats were anaesthetized with 5x10-3 mL/g BW 40% urethan subcutaneously. Piroxicam unabsorbed fraction was measured in a spectrophotometer at 353 nm, and then calculate Papp (Appear Permeability). The data obtained were calculated according to Kolmogorov-Smirnov and one-way ANAVA then continued by a t-test (95% confidence level). Tween 80 at 10% and 1.5% increased piroxicam absorption from the phosphate buffer pH 7.5 but decreased with Tween 80 2.0%. According to Papp, thevalues were 0.3929 cm/minute (1.5% Tween 80), 0.3507 cm/minute (1.0% Tween 80) and 0.2593 cm/minute (2.0% Tween 80) respectively. PEG 400 at 2.0 % rised piroxicam absorption from the phosphate buffer pH 7.5 but no with influence PEG 400 1.5 % concentration one way ANAVA. Abstract

Self-medication is defined as usage of drugs in community to relieve symptoms of minor illnesses without doctor intervention. Self-medication prevalence in Indonesia in 2004 is 24.1.% and in DIY province in 2005 is 87.73.%. The behavior of rational self-medication is probably influenced by sex, age, education level, occupation, income level, knowledge, and positive attitude toward self-medication. This was an observational research with cross sectional design. The number of respondent was 174, and the samples were chosen by multistage random sampling based on the villages number. Data was collected by using structured questionnaire and nonpartisipative observation. The data was analyzed by using independent sample t-test, Pearson correlation and multivariate analysis with multiple logistic regression.The result of the research showed that there was significant relationship between sex, age, level of knowledge, occupation, income level, knowledge and attitude with rational self-medication behavior. Multivariate analysis result showed that education level was themost influencing rational self-medication behavior at confidence level 95.%. There was significant relationship between knowledge, attitude, sex, age, level of knowledge, occupation and income level with rational self-medication behavior. Education level was the most influencing rational self-medication behavior.Key words: self-medication behavior, self-medication knowledge, self-medication attitude

Abstract

Visible spectrophotometric method can be used to determine cefadroxil in the pharmaceutical dosage form. The reaction of cefadroxil with the condensation product of 2 mol ethyl acetoacetate and 1 mol formaldehyde in acidic condition (pH 3,5) at 45°C for 20 minutes yielded yellow product giving absorption at 367 nm. The concentration of Cefadroxil was calculated by using standard curve Y = 1.6063 X + 0.1634, r = 0.9932, p = 0,001 and Vxo = 9,7%. Determination of cefadroxil capsule ”X” (562.4 mg/capsule) showed that the concentration of cefadroxil observed is 490.5 mg/capsule (CV = 0.89%) and recovery value is 102.88% (CV = 0.67%). Abstract

Areca catechu seed contains antioxydant substances, supposed to have anticancer property. This research therefore addressed to examine the inhibitory effect of Areca catechu seed ethanolic extract (EP) on proliferating breast cancer cells, MCF-7. Areca catechu seed ethanolic extract (EP) standardization was done according to the standard of BPOM. Areca catechu seed powder extraction was done using ethanol 96%. Cytotoxic assay – to get the value of IC50 and to prevent the cell proliferation (using doubling time assay) – was carried out by using MTT assay. Apoptosis observation was done by acrydine orange- etidium bromide staining method (double staining). The result showed that treatment with Areca catechu seed ethanolic extract (25-100 µg/m) for 48 h caused 13-84% growth inhibition (IC 5077 µg/mL) of the cells, while arecoline (ARE) treatment (10-500 µg/mL) showed 8-73% inhibition (IC50 180 µg/mL). The extract also inhibited cell proliferation and induced apoptosis. These results conclude that Areca catechu seed ethanolic extract (EP) possesses antiproliferative effect through growth inhibition and apoptosis induction.Key words:MCF-7, Areca catechu, antiproliferative

Abstract

2,5- bis - ( 4’-hydroxy-3’- methoxybenzylidene ) cyclopentanone com- pound was know have cytotoxic activity to Mieloma , Raji and HeLa cells better than curcumin. In effort to discovered new compound which have cytotoxic activity, it necessery to modify this compound by changing some group on it with lossing methoxy group, so was yielded two compounds, there were 2,5- bis - ( 4’-hydroxy benzylidene) cyclopentanone and 2,5-bis – ( 4-chlorobenzylidene ) cyclopentanone. The synthesis was done by reaction between 4-hydroxyand 4-chlorobenzaldehydes and cyclopentanone through Claisen-Schmidt condensation reaction catalyzed by sulfuric acid with methanol and 2-butanol as solven. The structure of product was characterized by spectra of Infrared and Massspectroscopy, then anti proliferation activity to HeLa cells counted base on IC50 values The results showed that 2,5- bis -(4’-hydroxy benzylidene) cyclopentanone (IC50 value : 0.656 mM) have higher toxicity than 2,5- bis -(4’-chlorobenzylidene) cyclopentanone (IC50value : 5.376 mM).Key words : Claisen-Schmidt condensation; 2,5- bis -(4’-hydroxy benzylidene) cyclopentanone; 2,5- bis -(4’-chloro-benzylidene) cyclopentanone; anti proliferative; HeLa cell.

Abstract

The purpose of this research was to describe the pattern of acute of respiratory tract infections in children of 0-59 month’s old in Puskesmas I Purwareja Klampok, Banjarnegara. The data was taken by retrospective method from medical record. The data collected was analysed by nonanalytic descriptive statistic method, and then compared them to standard of therapy provided by Indonesian Health Departement. The result showed that all of 120 patients were pneumonia acute respiratory tract infections, which were 55.8% boys and 44.2% girls. The antimicrobials used were Cotrimoxazol (86.7%) and amoxicillin (13.3%). The major product used in that puskesmas was syrup (91.7%), and tablet that delivered in the form of powder was 8.3%. The treatment using these antimicrobial at Puskesmas I Purwareja, Klampok, Banjarnegara was in accordance to standard of therapy provided by Indonesian Health Departement.
